BOUIN-HOLLANDE’s Fixative for IHC
Bouin-Hollande Fixing Solution is a modified form of Bouin Fixing Solution specifically optimized for immunohistochemistry (IHC). It provides effective fixation and preservation of tissue samples while preserving morphology and intracellular components without compromising antigenicity.
Bouin-Hollande Fixing Solution is composed of saturated aqueous picric acid, formalin, acetic acid and copper sulfate. The combination of these ingredients provides effective fixation and preservation of cellular and tissue structures while maintaining morphology and intracellular components. The solution also decalcifies and chelates iron and calcium ions, lyses erythrocytes, and enhances IHC visualizations through antigen unmasking.
This fixation solution is widely used in the examination of soft tissues such as testes, ovaries, and intestines because it provides excellent preservation of cytoplasmic details and visualization of extracellular matrix structures, such as collagen fibers and reticulin fibers. Fixation with Bouin-Hollande solution is particularly useful for IHC applications because it allows optimal preservation of antigenic structures required for antibody binding.
In immunohistochemistry, Bouin-Hollande fixation solution is advantageous because it allows gentler fixation that preserves the antigenicity of tissue samples while preserving the structural details and morphology of cells. This is critical for successful IHC analysis, where antibodies are used to detect and localize specific antigens within the tissue.
