Diastase solution 0.1%, consisting of diastase and pure water, is an essential tool in histology, especially when performing PAS diastase staining. Diastase is an enzyme complex that has the ability to break down polysaccharides, such as glycogen, into their monosaccharide or oligosaccharide components.

During PAS-Diastase staining, this solution is used to remove glycogen from tissue samples. The underlying process is an enzymatic reaction in which the diastase hydrolyzes the glycogen. The pure water serves as a solvent and allows the diastase to be evenly distributed throughout the sample.

With its use, more specific staining of structural polysaccharides and glycoproteins in tissue samples is achieved.
