Orange II solution (PAP 2b) is used in the second stage of staining and is responsible for staining eosinophilic structures, such as the cytoplasm of superficial and intermediate cells, in orange to pink hues. This coloration allows better differentiation and analysis of the various cell types and their structures in cytological preparations, thus facilitating diagnosis.
