Rhodanine for Copper Detection

Rhodanine is an organic derivative of thiocyanate and is used in histology as a specific dye for the detection of copper, particularly in the liver. The rhodanine method is commonly used to diagnose Wilson's disease, a genetic disorder that results in excessive copper storage in various organs.

In practice, the tissue sample is first immersed in a rhodanine solution. If copper is present in the tissue, it reacts with rhodanine to form a red-orange complex. This complex is visible under a microscope and shows the presence and distribution of copper in the tissue.

It is important to note that the rhodanine method is sensitive and can detect trace amounts of copper. Therefore, it should be used in conjunction with other diagnostic tests and clinical findings. The performance of this staining method requires specialized laboratory equipment and expertise.
