SCHAFFER’s Fixative (buffered Formaline & Methanol)

SCHAFFER solution, also known as formalin-methanol-K-Na-Ph buffer, is a fixing solution commonly used in histology and cytology. It consists of formalin, methanol, and a potassium sodium phosphate buffer (K-Na-Ph buffer).

Formalin, an aqueous solution of formaldehyde, is a widely used fixative that cross-links proteins and nucleic acids in tissue specimens, preserving their structure. Methanol serves as a preservative and dehydrating agent that removes water from tissue samples, helping to stabilize cell structures.
K-Na-Ph buffer is a buffer that maintains stable pH levels, ensuring the optimal working condition for fixation.
