Safranine for GRAM’s Staining

Safranin is an important component of Gram staining, an essential technique in microbiology used to classify bacteria based on the properties of their cell wall.

In Gram staining, safranin is used as a counterstain to crystal violet, the primary stain. After cells are first stained with crystal violet and then treated with alcohol or acetone, Gram-positive bacteria retain the violet staining, while Gram-negative bacteria lose it. Safranin, applied next, stains these previously decolorized Gram-negative bacteria red.
