Weigert Stock Solution B is the second component of the Weigert iron hematoxylin stain, a commonly used histological staining method for visualizing cell nuclei and other basophilic structures in tissue sections and cell preparations. Weigert staining is a modification of hematoxylin staining and uses iron compounds as oxidizing agents instead of other chemical mordants.

Weigert stock solution B contains an iron salt solution, such as ferric chloride solution, which serves as an oxidizing agent. Stock solution A consists of a mixture of hematoxylin, ethanol and water. To prepare the working solution for Weigert iron hematoxylin staining, the stock solutions A and B are mixed in a 1:1 ratio.

The mixture of the two stock solutions A and B is used to apply the hematoxylin to the tissue while providing the iron compounds as oxidizing agents to develop the black-brown color of the hematoxylin dye. This mixture is applied to the tissue to be stained to selectively stain cell nuclei and other basophilic structures.
Weigert iron hematoxylin staining is often used as part of more complex staining procedures, such as in trichrome staining or Van Gieson staining, to provide differential visualization of the various tissue components.

Verwendung:

Weigert Eisenhämatoxylin wird für Zellkernfärbung für die Histologie und Zytologie eingesetzt. Die Kernfärbungen in Abstrichpräparaten, Dünnschichtpräparaten, Gefrierschnitten und Paraffinschnitten ergeben sehr stabile Ergebnisse, die keine zusätzliche Differenzierung benötigen.Das Eisenhämatoxylin wird sehr oft bei Trichromfärbungen eingesetzt, bei denen mit sauren Farbstofflösungen gegengefärbt wird, z.B. Masson Goldner Trichrom (Artikel-Nr.: 12043). Das Reagenz wird jedoch auch allein als Färbemittel zur Darstellung von Tonofibrillen, Gliafasern, Kernteilungsfiguren, Mitochondrien und Markscheiden eingesetzt. Die Färbelösung ist ausschließlich für die professionelle Anwendung als in-vitro Diagnostikum im histologischen Labor vorgesehen.
