Methyl green stem B (acetate buffer, pH 4.8)
Methyl Green Stock Solution B is another histological staining solution that is used in combination with Methyl Green Pyronin Stock Solution A. Stock Solution B contains an acetate buffer with a pH of 4.8, which is used to maintain the pH in the optimal range for interaction between the dyes (methyl green and pyronine) and the nucleic acids (DNA and RNA) during staining.
Methyl green pyronine stain is used to visualize nucleic acids in tissue sections. Methyl green binds selectively to DNA and stains it green, while pyronine binds to RNA and stains it red. By using stock solution A and B, differential staining of DNA and RNA can be achieved.
The stain is applied in various histological and cell biology studies to visualize cell nuclei and nucleic acids. It can also be used to better detect and analyze cellular changes associated with various diseases.
