Safranin, alcoholic

Safranin is a synthetic cationic dye used in histology, cytology and microbiology. The alcoholic solution of safranin consists of safranin in a suitable alcohol, usually ethanol or methanol, as a solvent.

In histology, safranin is commonly used to stain cartilage, bone, and nucleic acids. It binds to acidic structures and stains them red or reddish brown. Safranin is also commonly used in plant biology to stain cell walls, plasmodesmata, chromoplasts and other plant cell structures.

In microbiology, safranin is an important component of Gram staining, a widely used method for classifying bacteria. After decolorization with alcohol or acetone-alcohol mixture, safranin is used as a counterstain to stain Gram-negative bacteria red, while Gram-positive bacteria remain purple.

The alcoholic safranin solution should be stored away from light and at room temperature to maintain its stability and staining power.
