Staining Kit: MOVAT’s Pentachrom after VERHOEFF
The Movat Pentachrome stain shows the following tissue components:
1. Elastic fibers: black (by Verhoeff elastin stain)
2. Collagen: yellow (by True Saffron - Safran du Gâtinais)
3. Basic substance (mucopolysaccharides): blue (by Alcian blue)
4. Muscle fibers: red (by Brilliant crocein acid fuchsin solution)
5. Erythrocytes: yellow-orange (by Brilliant crocein acid fuchsin solution)
The kit consists of different solutions used in a specific order to obtain the desired staining results. The staining is done with 2% phosphotungstic acid. The use of the Movat Pentachrome staining kit according to Verhoeff requires precise protocols and careful handling to ensure optimal staining results. This method is particularly useful for the analysis of vascular wall layers, especially in the study of atherosclerosis, vascular abnormalities, or inflammatory changes.
Cartilage tissue is stained blue-green to dark green by metachromatic effects. Collagenous tissue (light yellow) can be differentiated from young osteoid (dark yellow); depending on fixation, osteon structures (reddish) are labeled. Muscle tissue, cytoplasm (red) and cell nuclei (blue-black) are differentiated. Modification according to Verhöff allows differentiation of elastic fibers (black), myelin sheaths (black-brown) and more intense nuclear staining.

The Movat Pentachrome staining kit according to Verhoeff is a histological staining method for in vitro diagnostics that enables the simultaneous visualization of five different tissue components. It is particularly used for the analysis of vascular wall layers, cartilage and bone tissue, as well as for the investigation of atherosclerosis, vascular abnormalities and inflammatory changes.
