Staining Kit: PAS-Diastase
The staining kit: PAS-Diastase represents a balanced ensemble of components specifically composed for the detection of glycogen in tissue samples. Among them are Schiff's reagent, hematoxylin according to GILL - III, diastase solution 0.1 % and periodic acid 0.5 %.
The periodic acid is responsible for the oxidation of 1,2-glycols to aldehydes, which react in the subsequent step with the Schiff's reagent to fuchsin. This process stains the glycogen red. To enable more specific staining, the diastase solution is used. It breaks down glycogen in the tissue sample. The hematoxylin according to GILL - III serves as a counterstain.
