Staining Kit: PAPPENHEIM’s Staining (MAY GRUENWALD & GIEMSA)
The staining kit for Pappenheim staining (May-Grünwald & Giemsa) is frequently used in medical diagnostics and research. It consists of two main components: May-Grünwald solution and Giemsa solution.
The May-Grünwald solution serves as a pretreatment and fixes the cells. It contains methylene blue and eosin and stains the cells already slightly.
The Giemsa solution is a mixture of azure II, eosin and methylene blue and is used for the actual staining. It stains nuclei dark blue to purple and cytoplasm pink to red. Particular structures such as granulocyte granules or malaria parasites can take on different colors, allowing for differentiated diagnosis.
Pappenheim stain is a panoptic/panchromatic stain for blood smears because it can visualize basophilic, neutrophilic, and eosinophilic structures. This feature allows comprehensive visualization of the different cell types and their structures, which is of great importance for the diagnosis of blood diseases, infections and parasite infestations.
