Verwendung:
The staining kit for silver impregnation according to Bielschowsky is used for in vitro diagnostics in the context of histological examinations. It enables the visualization of neurofibrils, reticular and collagen fibers in prepared tissue samples. The procedure stains neurofibrils and reticular fibers black, while collagenous fibers take on a brown stain. The kit composition includes solutions such as potassium permanganate, potassium metabisulfite, silver nitrate, formalin, gold chloride, and sodium thiosulfate, which are used according to the Bielschowsky method to ensure effective staining and subsequent examination of reticulin fibers in histological specimens.
Prinzip:
The silver impregnation method according to Bielschowsky is a histological staining method for visualizing nerve cells, axons and neurofibrillary structures in tissue samples. In this process, complex formation and reduction lead to the attachment of silver salts to certain cellular components. Potassium permanganate is used to pretreat and oxidize the tissues, while potassium metabisulfite and silver nitrate lead to the formation and deposition of elemental silver on the target structures. Formalin fixes the tissue samples, while ammoniacal silver nitrate solution and gold chloride stabilize and intensify the staining. Sodium thiosulfate removes excess silver and reduces non-specific staining. This method is used in in vitro diagnostics for the investigation of neurological diseases.
